Resumen : Fondo: El factor de transcripción STAT3 tiene un importante efecto de inmunidad innata en la progresión del cáncer. Se determinó la regulación de STAT3 en la modulación del inmunofenotipo de macrófagos de M1 a M2 inducida por el sobrenadante de cultivo celular de la línea de próstata-cáncer PC3. Métodos: Monocitos: los macrófagos de donantes sanos se cultivaron en el sobrenadante de células PC3, proteínas de membrana y STAT3 intracitoplasmático y fosforilado, se midieron mediante citometría de flujo, mientras que las citoquinas y los factores de crecimiento se estudiaron mediante luminiscencia. La citotoxicidad y el óxido nítrico se evaluaron mediante ensayos colorimétricos. Resultados: El sobrenadante de células de tumor de próstata PC3 indujo eficazmente macrófagos hacia un perfil M2, y la expresión de STAT3 fosforilada en los monocitos-macrófagos aumentó notablemente, y se relacionó principalmente con IL-10. En el grupo de monocitos-macrófagos tratados con un inhibidor de STAT3, los macrófagos se indujeron hacia un fenotipo M1. Conclusiones: En este estudio, mostramos que el perfil de secreción de células de cáncer de próstata PC3 induce un cambio en el fenotipo de macrófagos de M1 a M2, y que el fenómeno está relacionado con la fosforilación del factor de transcripción STAT3 e IL-10.
Descripción : BACKGROUND: Transcription factor STAT3 has a prominent innate immunity effect on cancer progression. We determined the regulation of STAT3 in the immunophenotype modulation of macrophages from M1 into M2 induced by the cell-culture supernatant of the Prostate-Cancer line PC3. METHODS: Monocytes-macrophages from healthy donors were cultured in the supernatant of PC3 cells, membrane proteins, and intracytoplasmic and phosphorylated STAT3 were measured using flow cytometry, while cytokines and growth factors were studied using luminescence. Cytotoxicity and nitric oxide were evaluated via colorimetric assays. RESULTS: The supernatant of PC3 prostate-tumor cells effectively induced macrophages toward an M2 profile, and the expression of phosphorylated STAT3 in the monocytes-macrophages notably increased, and mainly related to IL-10. In the group of monocytes-macrophages treated with a STAT3 inhibitor, the macrophages were induced toward an M1 phenotype. CONCLUSIONS: In this study, we showed that the secretion profile of PC3 prostate-cancer cells induces a change in macrophage phenotype from M1 into M2, and that the phenomenon is related to phosphorylation of transcription factor STAT3 and IL-10.
