Resumen : Se probaron cinco protocolos de aislamiento de ADN genomico: Clarke et al., (1989), Istvan Nagy et al., (1998), Dellaporta et al., (1983), Saghai- Maroof et al., (1984)y el método de Clarke et al., (1989)modificado por Guillen et al., (2003). Todos los protocolos indicados se han utilizado con éxito en diferentes especies vegetales a excepción del de Clarke et al., (1989) modificado. En todos los procedimientos de aislamiento de ADN, se estandarizó la cantidad de tejido foliar utilizado.Con excepción del método propuesto por Saghai-Maroof et al., (1984)el tejido foliar fue colectado el día previo a su procesamiento y se almacenó a una temperatura de -20 °C. Los iniciadores microsatelites del tipo ISSRs (intersecuencias simples repetidas) empleados fueron siete para obtener un amplio numero de fragmentos de ADN amplificado por secuencias aleatorias de 77 colectas (231 individuos), de aguacate (Persea americana Mill.) raza mexicana existentes en el Banco de Germoplasma de INIFAP- CEFAP-Uruapan, Michoacan. El protocolo descrito por Clarke et al., (1969), modificado por Guillen et al., (2003), resulto ser un método viable para ser utilizado en aguacate. Los marcadores genéticos ISSRs fueron capaces de detectar altos porcentajes de polimorfismo desde un 82.28 a 95.39 %. Se formaron dos grandes grupos, el primero con 11 subgrupos y el segundo con tres subgrupos. Se encontraron materiales con un numero similar de bandas polimorficas, en el subgrupo la provenientes de las colectas 237 de Atlixco, Puebla y CXTC01 de Uruapan, Michoacan, las más disimiles fueron las colectas 532 de Atlixco, Puebla y 309 de Chilchota, Michoacan, pertenecientes al segundo grupo. Existieron algunas variaciones en el agrupamiento de los materiales de Persea americana Mill, La amplia diversidad de las colectas en el banco de germoplasma del CEFAP-Uruapan indico que no existieron materiales duplicados por lo que deben mantenerse para posteriores trabajos de mejoramiento genético.
