Resumen : Temefos es un plaguicida organofosforado usado ampliamente en las campañas de salud y fumigación urbana. A pesar del uso generalizado de este plaguicida, pocos estudios en la literatura han examinado su potencial genotóxico a bajas concentraciones. El objetivo de este estudio fue investigar si temefos tiene un efecto citotóxico, citostático o genotóxico en células HepG2. Para evaluar la citotoxicidad, se utilizó el ensayo colorimétrico MTT. Los efectos citóstatico y genotóxico se determinaron por el método propuesto por Eastmond y Tucker (1989) y por el ensayo de micronúcleos por bloqueo de citocinesis (MNBC) respectivamente. Se determinó la frecuencia de puentes nucleoplasmicos, gemaciones,células apoptóticas y necróticas. Los tratamientos con temefos (0.5- 10uM) durante 48 horas no afectaron la viabilidad celular, ni modificaron el indice de división nuclear (IN), sin embargo el tratamiento con temefos a 10 uM aumentó significativamente el numero de micronúcleos. Los resultados de este estudio sugieren que la biotransformación de temefos podría generar metabolitos genotóxicos en células HepG2.
Descripción : Temefos is an organophosphorus pesticide widely used in health campaigns and urban spraying. Despite the widespread use of this pesticide, few studies in the literature have examined its genotoxic potential at low concentrations. The objective of this study was to investigate if temefos has a cytotoxic, cytostatic or genotoxic effect in HepG2 cells. To evaluate the cytotoxicity, the MTT colorimetric assay was used. The cytostatic and genotoxic effects were determined by the method proposed by Eastmond and Tucker (1989) and by the micronucleus test by cytokinesis blockade (MNBC) respectively. The frequency of nucleoplasmic bridges, gemations, apoptotic and necrotic cells was determined. The treatments with temephos (0.5-10uM) during 48 hours did not affect the cell viability, nor did they modify the index of nuclear division (IN), however the treatment with temephos at 10 uM significantly increased the number of micronuclei. The results of this study suggest that the biotransformation of temephos could generate genotoxic metabolites in HepG2 cells.
